However, playing a tower defense game with things from Minecraft sounds pretty cool to me. Perhaps there could be special rounds, like an End or Nether round, where all of the mobs are from the respective dimensions.

   My general idea on my server would be to setup a path (say fences or just a ditch) with a mob spawner tower of some sort at one end and a player would stand at the other end. The player(s) would spend time building dispensers and filling with arrows and setup redstone to wire to a pressure plate at the end where the player would stand as well as to the doors to the mob spawner tower.
