Used generally by late 19c. for any mixture or preparation of unknown ingredients. Extension to "narcotic drug" is by 1889, from practice of smoking semi-liquid opium preparation.
The earliest known use of the verb dope is in the 1860s. OED's earliest evidence for dope is from 1868, in Putnam's Magazine. It is also recorded as a noun from the 1850s.
