Mifune had resistance and knowledge of Hanzo, but Hanzo beat the sannin without poison. The point of the question is that you have to think about whether or not current Jiraiya was as strong as the 2nd shinobi war sannin when they fought Hanzo.
Rewatching this episode made me realize that Hanzo wants the same thing as Mifune, Naruto, Nagato, he just takes a much different approach. He truly cared about one United shinobi world. The fight between Mizune and Hanzo felt very similar to Naruto’s meeting with Nagato.
