- Venom. Venom had to be at the top of the list, because he’s Venom, right? Sort of. Yes, he’s a great character and, with Harry at the wheel there’s a real note of tragedy there.
- Kraven the Hunter. Like Mysterio, the original Kraven the Hunter is a bit hammy, so Insomniac had its work cut out making him a genuine menace. Yes, he kills Scorpion and he’s downright terrifying, to the point where you wonder why he needs minions at all.
- Mr Negative. Mr Negative’s boss battle features more illusory/mindscape shenanigans, which is always fun. What’s so excellent about Martin Li aka Mr Negative is that he gets a shot at redemption — and takes it.
- Scream. Mechanically, Scream’s boss battle is fun but what really makes this symbiote-infected Mary Jane such a blast is the verbal barbs she dishes out during the fight.

- Mary Jane's Scream. Host. Scream. Mary Jane Watson. Shapeshifting, claws, enhanced physical abilities. Laura Bailey. Prior to the final act, Venom turns Mary Jane into an offspring, Scream.
- Venom. Host. Harry Osborn (and briefly, Peter Parker) Superhuman strength, regeneration, shapeshifting, hive mind. Tony Todd. After being extracted from Peter, the symbiote merges with Harry again.

- Kraven The Hunter. When you have a villain who can make you feel for other Spider-Man villains, you know you have a fantastic supervillain on your hands.
- Venom. Venom is truly the star of the show. Voiced by legendary horror icon Tony Todd, it doesn't get much better than that for this character. Well, actually, you also get to play as him later in the story when you're taking on Kraven, so that's a nice bonus for his character as well.
- Sandman. Sandman, aka Flint Marko, is the unexpected introductory boss fight in the sequel game. The fight is even more intense and cinematic than the Kingpin one in the first Spider-Man and Rhino in Miles Morales, showcasing the towering size Sandman can get and all his shapeshifting powers that allow him to make spikes from his arms and create minions.
- Mysterio. Mysterio is a pleasant surprise to see in Marvel's Spider-Man 2 and handled quite well. Even though he comes with a boss battle, he's not your main villain boss, and nor is he a true villain in the story.

- Venom. Harry Osborn. Right before Kraven is dealt with, Venom is born. The symbiote takes full control of Harry in the Oscorp Building which is where Venom becomes playable in the game.
- Mr. Negative. Martin Li. This was one of the two main villains in the first game. Mr. Negative returns in Marvel’s Spider-Man 2 as a prisoner before he is eventually broken out of The Raft.
- Mysterio. Quentin Beck. Mysterio is surprisingly a very sympathetic character because like so many villains in Marvel's Spider-Man 2, he, as Quentin, has changed.
- Kraven The Hunter. Sergei Kravinoff. Speaking of Kraven, he begins Marvel's Spider-Man 2 as the main antagonist although that will eventually shift. His lore changes between mediums, but in this game, Kraven is dying.
