The formation of double strand breaks (DSBs), the most lethal form of DNA damage, may be a physiological process that modifies chromatin organization and gene expression involved in information processing, learning, and memory and may be caused by normal brain activity.
We found that H 2 O 2 treatment of PC12 cells induces an increase in DNA-PK protein complex levels, along with an elevation of DNA damage, measured both by the formation of γΗ2ΑX foci, detected by immunofluorescence, and γH2AX levels detected by western blot analysis.

In this Commentary we present and discuss strength and weakness of several strategies for the direct cellular reprogramming from somatic cells to generate human brain cells which maintain age-related features.
